AIDS 개요

에이즈(AIDS)란, Acquired Immune Deficiency Syndrome의 약자로서 후천성면역결핍증 또는 인간면역결핍증으로 불리워집니다. AIDS는 원인 바이러스인 HIV의 감염에 의해 우리 몸속 질병에 대한 방어기능이 상실되는 병으로서, 초기증상으로는 현저한 체중감소, 원인모를 만성적 설사, 발열, 지속적인 마른기침, 질병 진행 시에는 피부염과 물집, 구강염, 식도염, 대상포진, 폐렴, 결핵, 카포시육아종 등 합병증으로 사망하게 됩니다.
즉, AIDS 란 면역기능이 저하되어, 건강한 인체 내에서는 활동이 억제되어 병을 유발하지 못하던 세균, 곰팡이, 바이러스, 기생충 등이 병원체로 재활하거나 새로운 균이 외부로부터 침입, 증식함으로써 발병하는 일련의 모든 증상을 총칭하는 말입니다.

* 전 세계 감염자 현황

HIV 감염에 대한 꾸준한 계몽활동, 효과적인 치료와 예방프로그램을 포함한 세계적인 노력은 꾸준히 지속되어 왔습니다. 하지만, 2017년 WHO (World Health Organization)의 발표 자료에 따르면, HIV에 감염된 인구는 이로 인한 사망자와 함께 지속적으로 증가하여 2016년 약 3천 7백만명에 달하는데 이 인구는 2000년 기준으로 20% 증가한 것으로1990년의 약 3배에 달하는 감염인구수를 보이고 있으며, 2016년에만 약 1백 8십만명의 새로운 HIV 감염 환자가 늘어나고 있으며 약 1백만 명이 HIV/AIDS로 인해 사망하였습니다.
세계 여러 지역에서 신규 감염자중 40%가 15세에서 24세 사이에 집중되어 있습니다. 전체 감염자 중 약 63%가 사하라 이남 지역의 아프리카에, 약 32%가 남아프리카 지역에 거주하고 있으며, AIDS로 사망한 인구 중 약 34%가 이 남아프리카 지역에 거주하고 있습니다. 동유럽 및 중앙아시아 지역에서의 AIDS 신규 감염자도 급속히 증가하고 있는 경향입니다.

* 국내 HIV 감염자 현황

질병관리본부의 2017년 8월 발행 "2016년 HIV/ADIS 신고 현황"에 따르면 2016년 한 해 1,199명이 신규로 신고되어 역학 조사가 시작된 1985년 이후 매년 신규 감염자가 꾸준히 증가되고 있습니다. 이로써 2016년 현재 HIV/AIDS 내국인은 11,439명의 감염자가 있는 것으로 파악되고 있고 성별로는 남자 92.8% (10,618명), 여자 7.2% (821명)으로 남성의 감염 비율이 월등히 높은 것으로 나타났습니다. 감염 경로는 성접촉(99%)이 대부분이며, 혈액제제에 의한 감염은 1995년, 수혈로 인한 감염은 2006년 이후 보고된 사례는 없는 것으로 나타났습니다. 그러나 공식적인 집계 외에도, 검사를 받지 않고 숨어 지내거나, 자신이 감염된 사실을 모른 채 살고 있는 집계 수치 이외의 감염자를 고려했을 때, 국내 HIV 감염자는 약 13,000 명 (7,500~42,000 명 범위)으로 추산하고 있습니다.

에이즈 시장 동향

1983년에 HIV가 AIDS의 원인균으로 밝혀진 이후 HIV/AIDS 백신 후보물질에 대한 전임상 및 임상 연구는 지속되었지만 상용화 된 백신은 아직까지 전무한 상태입니다. 최근에는 AIDS 백신 연구의 선두 주자인 Merck 사가 개발 중인 백신의 임상 2상 시험을 중단 하면서 이에 대한 여파로 AIDS 백신 연구자체가 주춤한 상태였으나, 미약하지만 효력을 보였던 태국의 임상 3상 시험 RV144의 여파로 중화항체를 중점적으로 공략하는 백신 후보 물질들의 연구 및 임상 시험이 주도적으로 진행되고 있습니다.
IAVI(International AIDS Vaccine Initiative) 통계에 따르면, 2017년 기준 현재 진행 중이거나 개시 예정인 치료 및 예방용 에이즈백신 임상시험은 대략 45여건으로 집계되고 있는데, 대부분 HIV의 일부 유전자를 이용한 DNA 또는 단백질을 이용하는 백신 및 중화항체를 분리, 배양하여 백신으로 이용하는 형태의 백신이 주류를 이루고 있습니다. 스마젠은 에이즈 백신인 SAV001 의 임상 2상 시험도 IAVI에 예정 중인 임상 시험으로 기재되어 소개 되어 있습니다.

강칠용 박사 연구의 특징

* Conventional Wisdom Approach

현재 대부분의 에이즈 백신 후보 물질들은 AIDS virus의 항원(gp120, gp41)만을 이용하여 예방용 백신을 위한 임상 시험을 개발하고 있습니다. 가장 선두 주자로 임상을 진행하고 있는 한 에이즈 백신의 경우, 최근 임상 3상 시험에서 미비한 예방 효과가 있는 것으로 발표 되었으나, 상용화 백신으로 개발을 위해서는 부족한 결과로 평가되고 있습니다. 이처럼 AIDS 백신은 HIV의 일부 항원만을 사용하는 것에는 효력이 한계가 있다는 것을 알 수 있었습니다. 한편, HIV가 아닌 다른 바이러스 연구에서는 바이러스 전체를 사용하여 백신을 만들어서 성공한 사례가 있고, 동물용 백신에서는 일반적이라는 점 등에 착안하여, 강칠용 박사는 AIDS 백신 연구에서는 최초로 HIV 전체를 이용하여 백신을 만들어 보자는 아이디어(Conventional wisdom)를 채택하였습니다.

연구 진행 경과 및 향후 일정

* 스마젠 에이즈 백신 동물 효력 검증 및 전임상 독성 시험

스마젠 에이즈 백신은 HIV 바이러스를 구성하는 전체 구조를 가지고 있고, 혁신적인 유전자 재조합 기술을 이용하였다는 점에서 다른 여타의 백신들과는 확연히 차별화 되고 있습니다. 전체 바이러스를 이용하는 사백신 기술은 다른 질병 분야에서는 백신으로써 이미 적용되어 온 '전래의 지혜'에 근거를 두고 있지만, HIV는 그 자체의 감염 위험성과 생산기술적 문제로 인하여 에이즈백신에는 적용하기가 어려웠습니다. 스마젠의 기술력과 다양한 전략은 HIV 전체 바이러스를 항원으로 이용하는 것이 가능하도록 하였습니다.
스마젠 에이즈 백신의 효력을 알아보기 위해 2005년에 미국 University of California, Davis (UC Davis)의 영장류 전문 실험센터에서 원숭이를 대상으로 한 면역원성 시험이 시행되었습니다. 그 결과, 에이즈를 예방할 수 있는 항체를 생성할 뿐 아니라, HIV에 감염된 세포를 치료할 수 있는 치료세포를 생성하는 것이 확인됨에 따라 예방 및 치료 효과를 동시에 보일 수 있을 것으로 평가 되었습니다.
캐나다 몬트리올에 위치한 전임상 시험기관인 Charles River Laboratories에서는 면역보조제를 선정하기 위한 작업으로 설치류인 랫드를 대상으로 다양한 면역보조제를 적용해서 면역원성을 확인하는 시험을 수행하였으며, 미국 Alabama의 Southern Research Institute에서는 원숭이에서의 안전성을 평가하는 GLP (Good Laboratory Practice) 독성 시험을 통해 안전성에 전혀 문제가 없음을 보여주었습니다.
현재, 스마젠은 임상 2상 및 3상 등 후기 임상 시험에 대한 안전성 자료를 확보하기 위한 원숭이에서의 동물 시험 및 전체 사독 백신 (SAV001)과 다양한 형태의 에이즈 백신 후보 물질의 병합 투여에 대한 효력 평가를 위한 동물 시험에 대한 설계 및 계획을 준비 중에 있습니다.

* 대량 백신 생산 및 임상 시험 물질 공급

세포를 이용한 체외 배양으로 HIV 전체 바이러스를 대량생산 하는 것은 고도의 기술력이 필요한 작업일 뿐만 아니라 생산자의 안전성에 대한 고려를 위해 Biosafety Level 3 (BSL3)의 GMP 조건에서 생산해야 하는데 이러한 조건을 모두 갖춘 위탁생산업체(CMO, Contract Manufacturing Organization)는 세계적으로도 매우 드문 실정입니다. 스마젠은 여러 업체와 접촉 하고 논의 한 후 2007년 미국 메릴랜드에 위치한 Omnia Biologics (Omnia)에서 임상시험 물질 생산을 하기로 결정하고 계약을 체결한 후, Master Virus Bank를 생산, 비임상시험을 위한 시료 생산을 성공적으로 완료 하였으며, 2010년 임상 1상 시험을 위한 시료 생산을 완료 하였습니다.
전체 HIV를 사독화 한 세계 최초의 에이즈 백신인 SAV001 는 세계 최초라는 기대감과 동시에 완벽히 바이러스를 사독화 시켰는가에 대한 검증이 매우 중요한 사항입니다. 스마젠은 세포 배양액으로부터 SAV001를 정제하는 과정에서 화학적 불활화 과정을 거쳐 바이러스를 사독화 시켰으며 이후 최종 생산 단계에서 감마 조사를 통한 물리적 불활화 과정을 통해 완벽한 사독화를 성공하였고, 임상시료 생산 과정에 따른 불활화 단계별 사독화 정도 및 안전성 영역 확보 수치를 별도로 취합하여 FDA에 제출하여 안전성에 대한 검증을 받았습니다.

스마젠은 향후 후기 임상시험을 위해서는 보다 안정적이며 대량의 시료를 생산할 수 있는 생산 공정에 대한 연구를 수행하여 임상 2상 시료 생산은 Bioreactor를 이용한 대량 세포 및 바이러스 배양을 통해 생산 scale을 대폭 향상 시켰으며, chromatography column purification 과정을 통해 세포기질 DNA 및 기타 불순물을 최대한 제거한 고 순도 시료를 만들어 낼 수 있는 질적 향상을 달성하였습니다. 이를 바탕으로 임상 2상 및 3상 시험 물질 대량생산을 개시하여 5,000 여명 분의 시료 생산을 위해 CSU의 BioMARC과 생산 공정 개발 및 GMP 물질 생산에 대한 계약을 체결하여 진행 중에 있습니다.

임상 시험 진행

* 임상 1상 안전성 평가 시험 (SAV CT01)

스마젠 에이즈 백신 (SAV001)의 첫 번째 임상시험(SAV CT 01)은 이미 HIV-1에 감염되어 있으면서 항레트로바이러스제(HAART)를 6개월 이상 복용한 이력이 있는 양성 피험자를 대상으로 하여 진행 하였습니다. 첫 임상 시험으로 SAV001의 인체 안전성을 확인하고 이와 함께 SAV001이 유도하는 면역원 증강 현상에 대해 관찰하였습니다. 2012년 3월 첫 환자를 시작으로 미국 캘리포니아에 위치한 4 곳의 병원 임상시설에서 수행하여, 2013년 9월, 총 33명 환자에 대한 투여 및 투여 후 1년 동안의 관찰기간이 완료되었습니다. 임상 시험 결과 스마젠 에이즈 백신에 의한 인체 독성 및 부작용은 없었고 백신으로의 효과를 기대해볼 수 있는 면역반응이 매우 고무적으로 증가되는 현상이 관찰되었습니다.
백신을 투여 받은 피험자들은 백신투여 이후 1년 기간 동안에 에이즈 예방에 결정적인 역할을 할 것으로 예상되는 gp120에 대해서 일부 환자는 8배까지 항체가가 증가하였고, HIV를 구성하는 대표적인 단백질인 p24의 경우 일부 환자에서 512배까지 항체가가 증가되는 결과가 관찰되었고, 형성된 항체는 관찰기간인 1년 동안 지속적으로 유지되었습니다. 또한 에이즈 백신의 효력을 유발하는 가장 중요한 요소인 중화항체 형성에 대하여, 스마젠 에이즈 백신의 단회 투여 후 중화항체가 유도 되었다고, 대상 임상시험환자 대부분이 HIV-1 B형인 북미의 감염자이었음에도 불구하고 B subtype뿐 아니라 A와 D subtype에 대한 중화항체까지 형성됨으로 광범위 중화항체의 유도를 보여 주었습니다. 이는 SAV001이 HIV 감염 예방의 효력이 있는 백신으로의 기대를 한층 더 높여 주는 결과 라고 분석되고 있습니다.
스마젠은 본 임상 1상 시험 피험자들의 백신 투여 전과 후의 혈액을 모두 채취하여 HIV의 RNA 염기서열을 분석하였고, SAV001 백신과 같은 바이러스가 존재하는 지를 평가하여, 혹시 단 하나의 바이러스가 살아 남아서 증식을 하여 HIV 감염을 초래 할 가능성에 대한 검증을 하였습니다. 즉, 피험자들의 혈액 내에 SAV001 백신 바이러스의 RNA 염기 서열이 존재하지 않는다면 이는 SAV001 백신 바이러스는 증식을 전혀 하지 않고 완전 사독화가 되었음을 증명하는 것입니다. 분석 결과 33명의 피험자 모두에서 SAV001 백신 바이러스의 염기 서열은 전혀 관찰되지 않았습니다. 이로써 전체 사독 HIV 백신의 안전성을 또 한 번 입증하였으며, 이미 보여 준 뛰어난 면역 반응 유도 결과와 함께 세계 최초로 효과적인 HIV/AIDS 백신 상용화를 위해 한 발짝 더 다가가는데 힘이 되고 있습니다.
임상 1상에서 얻은 중요한 결과물은 학술 논문으로 편집하여 의학 및 바이러스에 대한 전문가의 의견과 기본 연구 자료 및 임상관련 결과를 기재하는 전문 학술지인 Retrovirology에 게재되었습니다.

* 임상 2상 효력 평가 시험

스마젠 HIV/AIDS 백신인 SAV001의 최종 목표는 전세계 최초로 성공적인 HIV/AIDS 백신 개발을 완료하고 상용화 하는 것입니다. 이러한 목표를 달성하기 위하여, SAV001의 1차 임상시험에서 얻어진 결과를 바탕으로 정상인에서 HIV의 감염을 예방하는 예방용 백신의 개발과 더불어 HIV 양성 환자에서 HIV를 소멸시켜 치료하는 치료용 백신의 개발을 병행하여 진행하는 계획을 수립하고 있습니다.
이미 임상 1상 시험을 통해 기본적으로 SAV001의 인체 안전성을 확보하였고, 이 결과를 바탕으로 임상 2상 시험에서 동성애자 등 감염 가능성이 높은 고 위험자들을 대상으로 하여 반복 투여에 따른 예방 효과를 검증하는 시험을 계획 중입니다. 또한 이와 동시에 HIV 감염자에서 혈중 HIV 수치를 낮추고 기존 항바이러스제의 복용을 중단하여도 바이러스 수치를 유지 하는지 치료 효과를 보기 위한 임상시험을 실시할 예정입니다.
본 임상 2상 시험의 시놉시스를 포함한 물질 생산 계획 및 결과 등에 대한 FDA와의 사전 미팅인 Type C meeting을 2017년 10월 진행 하여 FDA로부터 별다른 이의 사항에 없음을 확인 받았습니다. 이에 스마젠은 임상 2상 시료 생산이 완료됨과 동시에 임상 2상 시험을 개시 할 수 있도록 개발 계획을 수립하여 진행하고 있습니다. 임상 2상 시험은 미국, 캐나다를 비롯한 북미를 중심으로 진행될 예정이며, 이후 남아프리카 공화국 및 태국 등으로의 지역 확대도 고려하고 있습니다.

Sumagen's HIV/AIDS vaccine, SAV001, is a novel and unique HIV/AIDS vaccine. Unlikely other HIV vaccine candidates that are using a part of virus structure or genome, SAV001 is a whole killed HIV antigen with the genetic modification of deleting nef gene to reduce the virulence and of replacing env gene to enhance the production rate. In addition, chemical (aldrithiol-2) and physical (r-irradiation) inactivation methods are applied in the production process. SAV001 is the first HIV vaccine candidate using whole killed HIV as the antigen, and Now, Sumagen believes that SAV001 will be the only promising HIV vaccine candidate.

Acquired Immune Deficiency Syndrome (AIDS) is a set of symptoms and infections resulting from damage to the human immune system caused by the human immunodeficiency virus (HIV). This condition progressively reduces the effectiveness of the immune system and leaves individuals susceptible to opportunistic infections and tumors. The loss of bodyweight, diarrhea, fever, and dry cough are known symptoms of the initial AIDS infection. Most patients die from opportunistic infections or malignancies associated with the progressive failure of the immune system.

The number of people living with HIV/AIDS has increased dramatically since the disease was first discovered in the early 1980s. The lengthy campaign and education to prevent HIV infection has steadily reduced the number of newly HIV infected population. However, according to report of WHO in 2017, the number of people living with HIV worldwide continued to grow by 2016, reaching an estimated 37 million and it was 20% increase comparing in 2000 and about 3-times higher than 1990. Approximately 1.8 million new HIV infections occurred in 2016 and 1.0 million died caused by HIV/AIDS
HIV/AIDS is the most life threatening infectious disease in the South African area, and is the fourth largest cause of mortality in the world. Nearly 40% of newly infected people are young adults between 15 to 24 years, 63% are living in sub-Saharan Africa. KCDC ((Korea Center for Disease Control and Prevention) published "HIV/AIDS Epidemic in Korea, 2016" that there were 1,199 new HIV infections in 2016. The HIV infections have been consistently increased since 1985, and the total HIV infected people in Korea are 11,439 (92.8% of Male, 7.2% of Female. Sexual contact was the major route of infection (99%) and there was no HIV transmission through the blood agents or transfusion since 1995.

Status of HIV/AIDS Vaccine

Since HIV has been defined as the causative virus for AIDS in 1983, there have been many clinical trials conducted up to now. However, there have been many failed HIV vaccine candidates which did show the proper efficacies in Phase II/III clinical trials but no licensed HIV vaccine has resulted. Vaccine development against HIV/AIDS continues to struggle with the question of which components of the human immune response confer protection against HIV infection. Recently Phase III clinical trial in Thailand, RV144 trial, showed the results with modest protection of HIV and the trend of HIV vaccine research is focusing on inducing the broad neutralizing antibodies for both prophylactic and therapeutic HIV vaccine.
According to IAVI (International AIDS Vaccine Initiative), around 45 clinical trials of HIV/AIDS vaccines are ongoing or planning in 2017. Most of vaccines are using a part of virus structure or genome as the antigen. Sumagen's HIV vaccine, SAV001 is introduced as the planed Phase II clinical trial in 2017 at IAVI.

What makes Dr.Kang's project

* Conventional Wisdom Approach

Recently, an AIDS vaccine candidate failed during Phase III trials. The said candidate was developed with only parts of HIV antigens, gp120 and gp41, which once again implied with greater certainty that chances are slim with those HIV vaccine candidates that are based on certain parts of HIV antigens. Whole virus-based vaccines are a well-known approach to vaccines, filed as conventional wisdom. Furthermore, without a promising vaccine trial until now and scientific knowledge that components of HIV induce the immune response in human body, experts are discussing that the only way to make an HIV/AIDS vaccine seems to be to use a whole virus as the vaccine antigen. Sumagen's SAV001 is in this context the first whole-killed virus based HIV vaccine, which makes Dr. Kang's research unique.

SAV001 has been genetically modified in two ways to generate: (1) the natural HIV-1 glycoprotein signal peptide sequence has been replaced with that of honeybee melittin to enhance the production rate, and (2) the nef gene has been deleted to reduce the virulence. In addition, chemical and physical inactivation methods are applied in the drug production process. For the chemical inactivation method, aldrithiol-2 (2,2'-dipyridyl disulfide, AT-2) is used. Followed by the chemical inactivation, gamma irradiation is used for further physical HIV inactivation

Development Status and Future Progress

* Preclinical Efficacy and Safety Evaluation of SAV001

What makes HIV vaccine's clinical trials difficult is the fact that there is no cross-infection between human HIV and animal immunodeficiency viruses. To overcome this barrier, Dr. Kang and University of California at Davis laboratories cooperated to make new viruses called SHIV (Simian-Human immunodeficiency Virus) having SIV (Simian Immunodeficiency Virus) to transmit into monkey bodies. This monkey immunogenicity test was completed in August 2005, and the result showed high immune responses, not only to cell mediated immune responses but also to human immune responses with SAV001. Subsequent immunogenicity tests were followed using rats and mice at Charles River Laboratories in Montreal Canada, which resulted in enhanced immune responses when SAV001 was injected into animal bodies combined with adjuvant.
The safety of SAV001 with and without adjuvant was tested and proven through the non-clinical GLP (Good Laboratory Practice) toxicity study using monkey at renowned CRO (Contract Research Organization), Southern Research Institute, Birmingham United States. The study design was based on FDA and WHO guidelines. The result was that there were no severe toxicities with SAV001 even with higher dosages and multiple injections than what would actually be anticipated in a real case.
Sumagen are planning additional immunogenicity studies and toxicity studies to preparing Phase II and Phase III clinical trials.

* Large Scale Manufacturing and Phase II Material Production

The mass production with whole HIV at in vitro system is not an easy process. Sumagen has struggled with the process development because mass production with whole HIV manufacturing was the first attempt. Furthermore it is recommended that HIV be handled in a Biosafety Level 3 (BSL3) facility under Good Manufacturing Practice (GMP) conditions. To find an appropriate manufacturing facility to satisfy the BSL 3, as well as GMP conditions has also been a huge challenge. Finally, Sumagen found Omnia Biologics in Maryland, USA and signed a contract for SAV001 production for Phase I clinical trial in November 2007. After that, Sumagen succeeded with the Master Virus Bank (MVB) with cGMP (current GMP) conditions and cGMP production was completed at 2010. The cGMP drug products were inactivated twice through the manufacturing processes. Chemical inactivation was performed by AT-2 in purification processes, and physical gamma-irradiated inactivation was conducted after filling the drug substances in vials.

For the scaled-up manufacturing, Sumagen hired an additional cGMP BSL3 contracted manufacturing organization (CMO), BioMARC in Colorado State University (CSU). CSU has been developing and researching the scaled-up cell cultural system and down-stream purification process using column chromatography. Sumagen has contracted for the production of phase II or III clinical materials up to 5,000 vials of drug products in the near future.

* Phase I Clinical Trial (SAV CT01)

The first-in-man study of SAV001 (phase I, SAV CT 01) to evaluate safety and tolerability has been completed with HIV-1 positive subjects in the United States. The first subject was injected at March, 2012 and 33 HIV-1 positive subjects have completed for administration of either SAV001 or placebo.
According to the results, there was no adverse event observed after SAV001 treatment. Safety assessment was the major purpose for phase I clinical trial of SAV001 because it was the first trial to administer the killed-whole HIV vaccine in human body and the data showed that no safety concern remained using SAV001 for further clinical trials. In addition to the safety evaluation, HIV-1 antibody detections have been performed through follow up periods. Data from HIV-1 antibody detection, the antibodies against p24 and p17 antigens increased as much as 512-fold after vaccination in SAV001 and SAV001 plus adjuvant treatment groups. Antibody against gp120 antigen increased 2~4-fold after vaccination in SAV001 plus adjuvant treatment group.
It is believed that a truly efficacious prophylactic HIV vaccine should be able to harness the highly desired broadly neutralizing antibody response. Although the primary objective of this Phase I human clinical trial was an evaluation of the safety of a killed whole-HIV-1 vaccine, Sumagen tested the levels of neutralizing antibodies in plasmas of the vaccinated and control participants in Phase I clinical trial. The results showed that approximately 50% of subjects in SAV001 elevated neutralizing antibody level against HIV-1 subtypes B, D, and A. Some subjects' sera contain high levels of neutralizing activities. It was interesting to observe that most of the subjects were infected with HIV-1 subtype B in our trial, subjects were able to neutralize not only subtype B but also subtypes A and D. These dramatic results from phase I clinical trial are encouraging for higher expectation of induction of immune response in the patients and promising for efficacy in further clinical trials.
The complete inactivation of HIV-1 to generate a killed whole-HIV-1 vaccine is the most critical step to develop a killed whole-virus vaccine. To confirm the genetic mutation and safety of SAV001 vaccine, we attempted to PCR amplifying the SAV001-specific viral RNA in the plasma of subjects. All sequence reads were participant-specific and we did not identify SAV001 viral sequence in the >70,000 sequence reads. These findings further confirm the lack of detectable SAV001 virus replication within subjects which means SAV001 vaccine is completely inactivated. * Phase II Clinical Trial for Efficacy

Sumagen's goal for SAV001 development is to bring the first commercialized HIV vaccine to the market. To reach this goal, we consider two separated parallel clinical development plans for the SAV001 vaccine. Sumagen approaches to the therapeutic HIV vaccine development to be shortening the development and commercialization period, to accumulate additional data, and to use its resources more efficiently. And in parallel with the therapeutic approach, the prophylactic vaccine design will be prepared and performed.
The Phase I clinical trial with SAV001 (SAV CT 01) showed that SAV001 was considered tolerable for use in humans and induced the high immune-responses including broadly neutralizing antibodies. Based on the data from the Phase 1 clinical trial, Sumagen designed the Phase II clinical trial to investigate the therapeutic activity of SAV001 upon analytic treatment interruption (ATI).
Sumagen conducted the Type C Meeting with US FDA to discuss the synopsis of Phase II clinical trial and the improved large scale manufacturing process in October 2017. The meeting went smoothly and FDA concurred with the study design of Phase II clinical trial on the dosing regimen and target population, and also gave useful comments and recommendations on the production process. Sumagen is ready to start the Phase II clinical trial as soon as the cGMP production is completed. The Phase II clinical trial will be conducted in North America and will be extended globally to places such as South Africa and Thailand.
